Description

 
Immunogenicity of a protein reflects its capacity to induce specific activation of helper T-cells. These activated T-cells are then able to stimulate the production of protein-specific antibodies by B-cells.

 

The protein-specific activation of T-cells can be measured in vitro, allowing reliable evaluation of the intrinsic immunogenicity of a protein.

Features


The assay relies on antigen presenting cells and helper T-cells. The cells are collected from healthy donors with diverse HLA phenotype, representative of the majority of expressed phenotype in population.

 

The specific activation of T-cells directed against a molecule is measured by ELISpot (Enzyme linked ImmunoSpot assay). Non-molecule specific activation is measured for each healthy donor.

 

For each donor, controls include two reference proteins, respectively known to be immunogenic and non-immunogenic, allowing ranking of your protein to these reference proteins.

 

The analysis can be performed simultaneously for up to 5 proteins on the same set of donors.

Starting material


Depending on the size, we require 1 to 5mg of your protein.
The protein should be as pure as possible and free from any endotoxin. 

 

Deliverables


A report including:

• QC summary (including HLA phenotype and characterisation of donor cells)
• The specific immune response of Human T-cells to your protein for a sample of donors
• The specific immune response of same Human T-cells to immunogenic and non-immunogenic reference proteins
• Ranking of all analysed proteins as highly, moderately, weakly or non-immunogenic
